چکیده

The gene POU5F1 (also known as an OCT4) belonging to POU protein family is composed of 5 exons that can create many isoforms by alternative splicing. Most isoform OCT4A expressed in types stem cells. Its nuclear localization and presence the DNA-binding domain enable it regulate expression genes are responsible for pluripotency blocking differentiation Other splicing products numerous OCT4B (OCT4B-OCT4B5) differ from cell absence domain. Majority OCT4 were discovered only recently there have not been clear evidences about their role cells yet. OCT4A, OCT4B1 tumorigenesis already explored but without definite results. aim our research analysis variants association with patient´s prognosis occurrence metastasis. For this we used fresh surgical specimens patients colorectal cancer isolated primary tumours (n=47) metastasis (n=31). Subsequently, total RNA was transcribed cDNA. Through quantitative PCR method using particular TaqMan assays specific ΔΔ Ct method, separately analyzed relative A, B, also all together. We found out tumour significantly lower than adjacent non-tumour tissue. On other hand, increased metastatic tumor compared observed experiment probe whereas neither B reason Our indicate play a pivotal exclude A cause metastasis, consider important identify transcript.

OCT4B modulates OCT4A expression as ceRNA in tumor cells.

OCT4 is an essential transcription factor for maintaining the self-renewal and the pluripotency of embryonic stem cells (ESCs). The human OCT4 gene can generate three mRNA isoforms (OCT4A, OCT4B and OCT4B1) by alternative splicing. OCT4A protein is a transcription factor for the stemness of ESCs, while the function of OCT4B isoforms remains unclear. Most types of cancer express a relatively low...

Increased Expression of Two Alternative Spliced Variants of CD1d Molecule in Human Gastric Cancer

Background: CD1d presents glycolipid antigens to invariant natural killer T (iNKT) cells. The role of CD1d in the development of peptic ulcer and gastric cancer has not been revealed, yet. Objective: To clarify the expression of alternatively spliced variants of CD1d in peptic ulcer and gastric cancer. Methods: Patients with dyspepsia were selected and divided into three groups of non-ulcer dys...

Increased Expression of PHGDH and Prognostic Significance in Colorectal Cancer

Phosphoglycerate dehydrogenase (PHGDH) plays an essential role in cancer-specific metabolic reprogramming. It has been reported as a putative metabolic oncogene in several types of human malignant tumors, such as breast cancer and melanoma. To date, PHGDH expression in colorectal cancer (CRC) as well as its association with clinicopathological characteristics and prognostic implication remain u...

Increased claudin-1 protein expression in hepatic metastatic lesions of colorectal cancer.

BACKGROUND The molecular and morphological alterations of the tight junctions in hepatic metastatic lesions are poorly understood. The possible involvement of claudin-1 (CL-1), which is one of the major tight junctional proteins, was investigated in the tumorigenesis of hepatic metastasis in patients with colorectal cancer (CRC). PATIENTS AND METHODS A total of 14 patients with hepatic metast...

CIP2A expression is increased in prostate cancer

BACKGROUND The CIP2A protein is a recently characterized oncoprotein which inhibits protein phosphatase 2A activity. Expression of CIP2A has been detected in several carcinomas, but its expression and significance in prostate cancer has not been examined so far. METHODS Expression of the CIP2A protein was studied using immunohistochemistry in prostate cancer (n = 59) and in benign prostatic h...
